What software would I need to create a mesh design like this for the edge lit acrylic?

V carve pro would be all you would need even cut 2d would do it

One of the best for that kind of thing without breaking the bank is Rhino.

What he is showing is a 2D cut just a outline no 3D is needed

It is a 2D cut, but the outline or vectors are derived from a 3D mesh of a model. Sure, you could draw it in 2D, but you would need a good 3D minds eye to design it.
